АННОТАЦИЯ / ABSTRACT
Цель
Определение сроков выведения остаточных количеств флорфеникола и его метаболитов в форме флорфениколамина из органов и тканей свиней после применения им ветеринарного препарата «Флорипрем 40».
Материалы и методы
Изучение сроков выведения остаточных количеств действующего вещества препарата проводили на базе свиноводческого комплекса в Калужской области. Для научно-исследовательской работы использовали 21 животное (20 опытных, 1 контрольное) в возрасте 86 дней с массой тела 44-49 кг. Препарат задавали поросятам в течение 7 дней групповым способом в смеси с комбикормом в дозе 5 кг препарата на 1 тонну комбикорма. Убой животных (по 5 голов на срок) провели через 8, 12, 14, 16 суток после последней дачи препарата. Образцы мышечной ткани (правое бедро), печени, почек и кожи с подкожной жировой клетчаткой были отобраны от каждого животного в индивидуальные пакеты. Содержание флорфеникола и его метаболитов в форме флорфениколамина в экстрактах образцов органов и тканей поросят определяли методом высокоэффективной жидкостной хроматографии, остаточные количества флорфеникола в органах и тканях животных – с помощью кислотного гидролиза. Исследование выполнено с помощью хромато-масс-спектрометрической системы Shimadzu LCMS-8050 (Япония). Анализ и обработка данных проведены с использованием программного пакета Shimadzu LabSolutions (Realtime Analysis, Postrun Analysis, Quant Browser) v.5.99 SP2 (Япония).
Результаты
Результаты изучения динамики выведения остаточных количеств флорфеникола и его метаболитов из органов и тканей свиней показали, что флорфениколамин аккумулируется в основном в печени и почках, незначительные уровни флорфениколамина обнаружены в мышцах свиней. Полученные данные свидетельствует о высокой степени адсорбции действующего вещества исследуемого препарата в органы и ткани свиней и длительном нахождении в организме. Флорфеникол и его метаболиты в тканях кожи с подкожно-жировой клетчаткой свиней не обнаружены. Оценка сроков выведения флорфеникола и его метаболитов из организма свиней, проведенная при помощи программы WT1.4, показала, что срок выведения действующего вещества составляет: в мышцах – 5 дней, в печени – 14 дней, в почках – 10 дней.
Заключение
Употребление в пищу продуктов животного происхождения, полученных после курсового перорального применения препарата «Флорипрем 40» на свиньях, не представляет опасности для здоровья человека спустя 14 суток после окончания применения препарата.
Purpose
Determination of the period of elimination of residual amounts of florfenicol and its metabolites in the form of florfenicolamine from the organs and tissues of pigs after the use of the veterinary drug "Floriprem 40".
Materials and Methods
The study of the period of elimination of residual amounts of the active substance of the drug was carried out on the basis of a pig-breeding complex in the Kaluga region. For research work, 21 animals (20 experimental, 1 control) were used at the age of 86 days with a body weight of 44-49 kg. The drug was given to piglets during 7 days in a group method in a mixture with compound feed at a dose of 5 kg of the drug per 1 ton of feed. The slaughter of animals (5 animals per term) was carried out 8, 12, 14, 16 days after the last administration of the drug. Samples of muscle tissue (right thigh), liver, kidneys and skin with subcutaneous and fatty cellulose were collected from each animal in individual bags. The content of florfenicol and its metabolites in the form of florfenicolamine in extracts of samples of organs and tissues of piglets was determined by high-performance liquid chromatography; residual amounts of florfenicol in organs and tissues of animals were determined using acid hydrolysis. The study was carried out using a Shimadzu LCMS-8050 chromatography-mass spectrometry system (Japan). Data analysis and processing were carried out using the Shimadzu LabSolutions software package (Realtime Analysis, Postrun Analysis, Quant Browser) v.5.99 SP2 (Japan).
Results
The results of a study of the dynamics of excretion of residual amounts of florfenicol and its metabolites from organs and tissues of pigs showed that florfenicolamine accumulates mainly in the liver and kidneys; insignificant levels of florfenicolamine were found in the muscles of pigs. The data obtained indicate a high degree of adsorption of the active substance of the test drug into the organs and tissues of pigs and a long-term presence in the body. Florfenicol and its metabolites were not detected in skin tissues with subcutaneous fat of pigs. An assessment of the elimination time of florfenicol and its metabolites from the body of pigs, carried out using the WT1.4 program, showed that the elimination period of the active substance is: in the muscles – 5 days, in the liver – 14 days, in the kidneys – 10 days.
Conclusion
Consumption of animal products obtained after a course of oral administration of the drug "Floriprem 40" in pigs does not pose a danger to human health 14 days after the end of the drug.
